Indoxyl-β-D-glucoside (also known as 3-indoxyl-β-D-glucopyranoside) is a chromogenic substrate used to detect, identify, and enumerate microorganisms that produce the enzyme β-glucosidase. Upon hydrolysis by β-glucosidase, the substrate releases an indoxyl group that oxidizes to form an insoluble indigo dye, producing a distinct blue to blue-green color in β-glucosidase-positive colonies. It is commonly incorporated into culture media and membrane filtration assays to aid screening for β-glucosidase-positive organisms, including enterococci and C. perfringens.
